Background

Alpha2-Macroglobulinisthelargestmajornonimmunoglobulinproteininplasma.Thealpha2-macroglobulinmoleculeissynthesizedmainlyinliver,butalsolocallybymacrophages,fibroblasts,andadrenocorticalcells.Alpha2macroglobulinactsasanantiproteaseandisabletoinactivateanenormousvarietyofproteinases.Itfunctionsasaninhibitoroffibrinolysisbyinhibitingplasminandkallikrein.Itfunctionsasaninhibitorofcoagulationbyinhibitingthrombin.Alpha2-macroglobulinmayactasacarrierproteinbecauseitalsobindstonumerousgrowthfactorsandcytokines,suchasplatelet-derivedgrowthfactor,basicfibroblastgrowthfactor,TGF-ß,insulin,andIL-1ß.Isabletoinhibitallfourclassesofproteinasesbyaunique"trapping"mechanism.Thisproteinhasapeptidestretch,calledthe"baitregion"whichcontainsspecificcleavagesitesfordifferentproteinases.Whenaproteinasecleavesthebaitregion,aconformationalchangeisinducedintheproteinwhichtrapstheproteinase.Theentrappedenzymeremainsactiveagainstlowmolecularweightsubstrates(activityagainsthighmolecularweightsubstratesisgreatlyreduced).Followingcleavageinthebaitregionathioesterbondishydrolyzedandmediatesthecovalentbindingoftheproteintotheproteinase.
