Background

Alpha-2-MacroglobulindetectsMacroglobulin.Alpha-2-MACROGLOBULINisalargeplasmaproteinfoundintheblood.Itisproducedbytheliver,andisamajorcomponentofthealpha-2bandinproteinelectrophoresis.Alpha2-Macroglobulinisthelargestmajornonimmunoglobulinproteininplasma.Thealpha2-macroglobulinmoleculeissynthesizedmainlyinliver,butalsolocallybymacrophages,fibroblasts,andadrenocorticalcells.Alpha2macroglobulinactsasanantiproteaseandisabletoinactivateanenormousvarietyofproteinases.Itfunctionsasaninhibitoroffibrinolysisbyinhibitingplasminandkallikrein.Itfunctionsasaninhibitorofcoagulationbyinhibitingthrombin.Alpha2-macroglobulinmayactasacarrierproteinbecauseitalsobindstonumerousgrowthfactorsandcytokines,suchasplatelet-derivedgrowthfactor,basicfibroblastgrowthfactor,TGF-ß,insulin,andIL-1ß.Nospecificdeficiencywithassociateddiseasehasbeenrecognized,andnodiseasestateisattributedtolowconcentrationsofAlpha2macroglobulin.Theconcentrationofalpha2macroglobulinrises10-foldormoreinthenephroticsyndromewhenotherlowermolecularweightproteinsarelostintheurine.Thelossofalpha2macroglobulinintourineispreventedbyitslargesize.Thenetresultisthatalpha2macroglobulinreachesserumlevelsequaltoorgreaterthanthoseofalbumininthenephroticsyndrome,whichhastheeffectofmaintainingoncoticpressure.Antialpha-2-macroglobulinisidealforinvestigatorsinvolvedinserumcomponentproteinresearch.
