Thisantibodyisdesigned,produced,andvalidatedaspartofacollaborationbetweenRocklandandtheNationalCancerInstitute(NCI)andissuitableforCancer,ImmunologyandNuclearSignalingresearch.TheArfGAPsareafamilyofproteinsencodedby31genesinhumans.ThefunctionoftheArfGAPsistoregulatethesmallGproteinArf,aregulatorofmembranetrafficandactinCytoskeleton.TheArfGAPshavebeenfoundtobekeyregulatorsofcellularbehaviorsthatinvolvecoordinatedactinandmembraneremodeling,includingproteinsecretionandmigration.SeveralArfGAPsareassociatedwithcancercellinvasionandmetastasis,andtheArfpathwayhasbeenfoundtobeaffectedinanumberofgeneticdiseases.AlthoughtheimportanceoftheArfGAPsandtheArfpathwayforcellularphysiologyiswellrecognized,themolecularbasisforthefunctionoftheseproteinshasnotbeenestablished.Reagentsforthestudies,especiallyantibodiesspecificforparticularmembersoftheArfGAPfamilythatcanbeusedforimmunoblottingandimmunofluorescence,arenotavailable.ArfGAP3isoneofthreeArfGAPsthoughttobespecificallyassociatedwiththeGolgiapparatus;however,localizationhasonlybeendoneforectopicallyexpressedrecombinantproteinandcellularfunctionisstillnotestablished.Indeed,thereisagrowingcontroversyaboutthefunctionofArfGAP3togetherwithArfGAP1andArfGAP2.Antibodiessuitableforimmunoprecipitation,immunoblottingandimmunofluorescencewouldbeofgreatvalueinaddressingthecontroversyandfordiscoveringthepotentialroleofArfGAP3insecretionofproteins,suchasgrowthfactors,andtraffickingofotherproteinstothecellsurface,suchasgrowthfactorreceptorsandcelladhesionmolecules.
